MagAttract 96 Miniprep System

The MagAttract™ 96 Miniprep System is designed for state-of-the-art, fully automated, high-throughput plasmid minipreps, and combines the speed and efficiency of silica-based DNA purification with the convenient handling offered by magnetic beads. The high-purity DNA obtained in the MagAttract procedure is suited for use in applications such as automated sequencing. Principle

The MagAttract 96 Miniprep procedure combines an easy-to-automate magnetic-silica¨Cbead technology with a novel plasmid purification method. The unique chemistry used in the MagAttract procedure enables purification of plasmid DNA directly from crude lysates without the need for a lysate clearing step, allowing walk-away automation of the entire purification procedure.

Procedure

Cultivation and lysis are carried out in 96-well blocks, after which MagAttract Suspension is added to the crude lysate. Under the optimized buffer conditions, plasmid DNA selectively binds to the surface of the magnetic silica beads, while genomic DNA and cellular debris stay in solution. The bead-containing lysates are transferred to 96-well round-well blocks for the washing procedure. Pure plasmid DNA is then eluted using a low-salt buffer (provided) into 96-well microplates and is ready for use in downstream applications. The MagAttract 96 Miniprep procedure gives reproducible yields of up to 3 µg DNA from 1.25 ml E. coli culture.

Applications

The high-quality DNA obtained using the MagAttract Miniprep procedure is suitable for use in automated fluorescent sequencing applications, including capillary sequencing.

MagAttract technology is also used by the BioRobot HT Plasmid System, designed for walkaway purification and reaction setup for genomics projects.

The MagAttract 96 Miniprep Kit is designed for fully automated high-throughput plasmid minipreps. It is optimized for use on the BioRobot 8000, but is flexible enough to be used on any robotic workstation. The kit provides MagAttract Suspension (Miniprep) and buffers for 24 x 96 minipreps, yielding high-purity plasmid DNA (see figure "Long Read-Lengths Using the MagAttract 96 Miniprep System"). Accessories, such as multiwell blocks and microplates, can be purchased separately from QIAGEN to provide maximum flexibility in automation setup.

Long Read-Lengths Using the MagAttract 96 Miniprep System


Phred 20 analysis of 1722 shotgun-clone sequences from plasmid DNA isolated for a bacterial genome project. Clones were prepared using the MagAttract 96 Miniprep Kit, sequenced using BigDye Terminator chemistry, and analyzed using an ABI PRISM 3700 DNA Analyzer with POP-5 gel matrix. The analysis shows the percentage of sequencing reads, which were calculated using sequential windows of 10 bases.
