PCR products generated using Taq or other non-proofreading DNA polymerases have A overhangs on their 3'-ends. Using the pre-linearized pQE-30 UA vector (which has U overhangs on its 3' ends) and an optimized Ligation Master Mix, PCR products can be inserted into the expression vector with high stringency and efficiency. In contrast to common expression vectors, the need for restriction digestion of the vector or insert, primers with built-in restriction sites, or specifically designed adapters is eliminated; resulting in a much more efficient and robust cloning procedure. PCR products obtained using proofreading DNA polymerases can also be used for UA cloning after the addition of a 3'-end A overhang.
