For cleavage of 6xHis tags from proteins containing a Factor Xa Protease recognition site
Factor Xa Protease is a site-specific endoprotease that preferentially cleaves the C-terminal peptide bond of the recognition sequence Ile-Glu-Gly-Arg. The pQE-30 Xa vector encodes a 6xHis tag followed by this recognition sequence. By treating a purified, 6xHis-tagged protein expressed using the pQE-30 Xa vector with Factor Xa Protease, it is possible to obtain the protein free of vector-encoded amino acids. After 6xHis-tag cleavage, Factor Xa Protease is removed in a batch procedure using Xa Removal Resin.
