For use in combination with TAGZyme DAPase Enzyme for the removal of 6xHis tags from proteins expressed using TAGZyme pQE vectors
The recombinant TAGZyme Qcyclase and pGAPase Enzymes are used in conjunction with TAGZyme DAPase Enzyme for the removal of 6xHis tags from proteins expressed using the TAGZyme pQE-1 vector. Both Qcyclase Enzyme and pGAPase Enzyme carry a C-terminal 6xHis tag, allowing their removal from reaction mixtures by subtractive IMAC. They are used for processing of proteins that contain an inserted glutamine residue. The glutamine residue serves as a DAPase stop point when converted to pyroglutamate in the presence of excess Qcyclase Enzyme. After removal of DAPase Enzyme and Qcyclase Enzyme by subtractive IMAC, the pyroglutamate residue at the N-terminus of the target protein is removed by the action of pGAPase Enzyme. pGAPase Enzyme is removed by a second round of IMAC, allowing pure, detagged target protein to be recovered in the flow-through fraction.
