The TagĦ¤100 Antibody detects proteins expressed from the pQE-100 DoubleTag Vector. These proteins contain an N-terminal 6xHis tag and a C-terminal TagĦ¤100 that allow oriented binding of the protein to Ni-NTA HisSorb Plates or Ni-NTA Magnetic Beads via their 6xHis tags, and detection of the well-exposed TagĦ¤100 using the TagĦ¤100 Antibody (see figure "ELISA using Ni-NTA HisSorb Strips and TagĦ¤100 Antibody"). This system is ideal for ELISA procedures, for example for drug screening or accurate protein quantitation.

ELISA using Ni-NTA HisSorb Strips and TagĦ¤100 Antibody


Thioredoxin tagged with 6xHis at the N-terminus and with the TagĦ¤100 at the C-terminus was diluted in 0.2% BSA in PBS, pH 7.2, applied to Ni-NTA HisSorb Strips either with () or without () 25 µg/ml NIH/3T3 lysate protein and incubated for 1.5 h at room temperature (RT). The amount of immobilized protein was determined by incubating with the TagĦ¤100 Antibody for 2 h at RT followed by peroxidase-conjugated anti-mouse IgG secondary antibody for 1 h at RT and detected using o-phenylenediamine as substrate.
