The DNeasy 96 Plant Kit provides high-throughput isolation of high-quality DNA from 96 or 192 samples of plant leaf tissue in less than 2 hours. The simple 96-well procedure provides highly reproducible yields of total cellular DNA (see figures "Uniformity of DNA Yields from 96 Wheat Samples" and "Reproducible DNA Yield and Quality"). The streamlined DNeasy procedure and convenient 96-well format (see flowchart "DNeasy Plant Procedure and DNeasy 96 Plant Procedure") eliminate tedious labeling, one-at-a-time pipetting, and DNA resuspension steps, and help minimize cross-contamination (see figure "Reproducible DNA Yield and Quality"). Polysaccharides, proteins, and enzyme inhibitors are efficiently removed, making DNeasy purified DNA suitable for use in all downstream applications, such as PCR (see figure "Reliable PCR from Different Plant Species"), AFLP (see figure "AFLP Analysis of Eucalyptus DNA"), RAPD, and Southern blotting. Processing of DNeasy 96-well plates requires use of the QIAGEN 96-Well-Plate Centrifugation System.

Genomic DNA was purified from 2 eucalyptus trees (first 2 lanes) and their progeny (remaining lanes). After digestion with EcoRI and MseI, adapters were ligated to the ends of the fragments and adapter-specific primers were used to amplify AFLP fragments in 2 rounds of PCR. Uniformity of DNA Yields from 96 Wheat Samples

96 leaf samples were collected from a batch of wheat plants grown under identical conditions. 50 mg samples were disrupted in liquid nitrogen and DNA was isolated using the DNeasy 96 Plant procedure. |
