The DNeasy Tissue system is designed for fast and easy purification of total cellular DNA (genomic, mitochondrial, viral) from a wide range of biological samples including animal tissues, cultured cells, blood, yeast, and bacteria. DNeasy purified DNA is suitable for all PCR and Southern-blotting applications. DNeasy sample preparation technology is fully licensed, allowing DNeasy purified nucleic acids to be used in any molecular assay or other downstream application without risk of patent infringement.

The DNeasy Tissue system uses advanced silica-gel¨Cmembrane technology for rapid and efficient isolation of total cellular DNA without phenol, chloroform, or ethanol precipitation. Mechanical homogenization is not necessary since tissues are directly lysed by Proteinase K, reducing hands-on time. The buffer system is optimized to allow selective binding of DNA to the DNeasy membrane. Simple centrifugation processing completely removes contaminants and enzyme inhibitors such as proteins, divalent cations, and secondary metabolites.
Samples are lysed using the provided QIAGEN Proteinase K. Lysates are loaded onto DNeasy spin columns or a 96-well plate (see flowchart below). During a brief spin the DNA selectively binds to the silica-gel membrane while contaminants pass through. Remaining contaminants and enzyme inhibitors are removed in two efficient wash steps. Pure DNA is then eluted in water or low-salt buffer, ready for use.
