The QIAwell multiwell system is designed for high-throughput preparation of up to 20 µg ultrapure plasmid, cosmid, BAC, PAC, or P1 DNA. QIAwell purified DNA is ideal for use in all applications, especially sensitive applications such as small-scale, high-throughput transfection and reliable long-range automated fluorescent sequencing. The QIAwell system is a purification system based on patented QIAGEN anion-exchange technology and optimized for high-throughput vacuum-driven processing. QIAwell technology is available in 8-well-strip format for use on QIAvac 6S or in 96-well-plate format for use on QIAvac 96. QIAwell Kits provide different combinations of three component modules to match varied purification needs. Use of all three modules enables the procedure to be streamlined and automated on BioRobot Systems.
QIAfilter
modules (yellow) replace time-consuming centrifugation for clearing of
bacterial alkaline lysates. Rapid filtration through the special asymmetric
filter unit removes all denatured and precipitated cellular components in just 2
minutes. Particle-free filtrates flow directly into the QIAwell module.
QIAwell modules (clear) constitute the core of the QIAwell system where DNA purification takes place. The QIAwell system unites QIAGEN Anion-Exchange Resin with Empore membrane technology from 3M. QIAGEN Resin particles are enmeshed in an inert PTFE (polytetrafluoroethylene) matrix, combining the high capacity and selectivity of QIAGEN anion-exchange purification with the handling advantages of a membrane.
QIAprep modules (blue) desalt and concentrate purified DNA without precipitation. DNA eluted from the QIAwell module binds to the silica-gel membrane in the QIAprep module under highsalt buffer conditions. Ultrapure DNA is then eluted from the QIAprep membrane in the low-salt elution buffer provided at concentrations of 150¨C200 ng/µl, and is ready to use in any downstream application.
Bacterial cultures are lysed and the lysates are cleared by filtration through the QIAfilter module (see flowchart "QIAwell Plasmid Purification"). Plasmid DNA from the cleared lysates is selectively bound to the QIAwell membrane and contaminants are washed away. Ultrapure plasmid DNA is eluted in high-salt buffer, and is then concentrated and desalted either on the QIAprep module or by isopropanol precipitation.
