Principle

Operon¡¯s unique HTS platform has a higher capacity than any other available system (see figure "DNA Synthesis Platforms"), allowing unprecedented scale-up and speed. With a very attractive price, HTS oligos offer all high-volume genomics customers a cost-effective solution to their oligo needs.

In addition to the HTS system, our original flexible synthesis platform is optimized for synthesis of high-quality modified oligos. Our bar-coded production system captures real-time diagnostic information for each oligonucleotide, and any measurement that fails to meet our quality-control standards triggers an immediate resynthesis. To further ensure quality, randomly selected primers are analyzed by HPLC, PAGE, capillary electrophoresis, and MALDI-TOF mass spectrometry.

Operon HTS oligos are supplied in 96-well plates. Each well contains 200 µl of a 50 µM solution. Please contact your local ">customer service department if you require nonstandard concentrations or special aliquotting.

Dual-labeled probes are single-stranded oligonucleotides labeled with both a fluorophore and a quencher and are designed to hybridize to specific target sequences. Operon offers several types of dual-labeled probes, all adopting different principles of operation but sharing the same basic concept. Standard dual-labeled probes typically contain TAMRA at the 3' end and 6-FAM, HEX, or TET as a 5' reporter dye. These probes are commonly used with quantitative, real-time thermal cyclers, such as ABI. Sequence Detection Systems (ABI GeneAmp 5700 and ABI PRISM 7700 and 7900HT) and the LightCycler System. During the extension phase of the PCR, the 5' ¨C 3' exonuclease activity of Taq DNA polymerase cleaves the fluorophore from the quencher. This results in detectable fluorescence that is directly proportional to the amount of accumulated target.

Black Hole Quenchers (BHQ) are a new class of high-efficiency dark quenchers. The BHQ family was developed to provide excellent spectral overlap over the entire range of commonly used reporter dyes. They can be used in quantitative PCR, mutation detection, and allele determination.

Molecular Beacons are hairpin-shaped oligonucleotides with a 3' dabcyl quencher and a 5' reporter. When the probe finds its target, the hairpin loop opens and generates a fluorescent signal (see figure "Molecular Beacon Function"). The overall signal is proportional to the amount of target. Molecular Beacons can be used for quantitative PCR and to distinguish single nucleotide sequences for SNP and allele analysis.
