Principle

Repeated regions, GC content, and codon deviation may all affect the synthesis and function of a gene. Gene expression can be optimized by eliminating rare codons and utilizing optimal codons for your host or for multiple host systems. In addition, genes can be optimized by removing secondary structure, adjusting GC content, and adding or removing restriction sites.

After you have approved the final gene sequence, the GeneOp system is used to design oligonucleotides that minimize the incidence of direct repeats and other unwanted secondary structures that may hinder assembly or expression of the gene. After synthesis of the oligonucleotides, each duplex DNA product is carefully assembled by proprietary annealing and ligation reactions. The assembled gene is cloned, and the sequence is verified by complete bidirectional sequencing. The final product is delivered as lyophilized DNA cloned in a standard plasmid.
