生物通报道，哈佛大学生物化学系与分子药理学系的科学家近期在膜蛋白研究方面取得新进展，相关成果文章Reconstitution of Outer Membrane Protein Assembly from Purified Components公布在最新一期的Science杂志上。

 

文章通讯作者是来自哈佛大学的Daniel Kahne教授，他带领的研究小组从事细菌细胞壁生物合成以及抗生素的分子生物机制研究多年。

 

β-Barrel膜蛋白位于革兰氏阴性细菌、线粒体和叶绿体中，这种膜蛋白由高度保守和复杂的多蛋白复合体组成。β-Barrel膜蛋白的分子结构、折叠结构以及与酶作用的部位的结构机制却鲜少为人所知。研究β-Barrel膜蛋白的瓶颈在于，β-Barrel膜蛋白很难在一个生物化学体系重重新构建起来。

 

为了解决这一难题，Daniel Kahne教授研究小组将大肠杆菌E.coli的膜蛋白提取出来，并且重组了β-Barrel膜蛋白，并在蛋白磷脂体内重组，研究人员使用已有的酶来探索β-Barrel膜蛋白的折叠结构。

 

结果发现，β-Barrel膜蛋白的折叠是在一个没有能量的环境下进行的，其中一种可溶性的伴侣分子是β-Barrel膜蛋白折叠的关键因素。

膜蛋白

膜蛋白是指能够结合或整合到细胞或细胞器的膜上的蛋白质的总称。而细胞中一半以上的蛋白质可以与膜以不同形式结合。根据与膜结合强度的不同，膜蛋白可以被分为两类：外周膜蛋白和内在膜蛋白。

内在膜蛋白是整合于膜上的蛋白质，总是与膜结合在一起。可以定义为需要通过人工加入去垢剂（如SDS或Triton X-100）或其他非极性溶剂才能够从膜中分离出来的蛋白质。内在膜蛋白还可以根据与双分子膜之间结合关系的差异细分为：

跨膜蛋白，顾名思义即跨越膜的两端的蛋白质，其跨膜部分为β桶或α螺旋结构；

单向内在膜蛋白，其只从一个方向（膜外或膜内）与膜结合，虽然部分插入膜中，但不跨膜。

β-Barrel membrane proteins in Gram-negative bacteria, mitochondria, and chloroplasts are assembled by highly conserved multiprotein complexes. The mechanism by which these molecular machines fold and insert their substrates is poorly understood. It has not been possible to dissect the folding and insertion pathway because the process has not been reproduced in a biochemical system. We purified the components that fold and insert E. coli outer membrane proteins and reconstituted β-barrel protein assembly in proteoliposomes using the enzymatic activity of a protein substrate to report on its folding state. The assembly of this protein occurred without an energy source, but required a soluble chaperone in addition to the multiprotein assembly complex.
