我国科学家解析人源蛋白复合物三维结构

Structure of Human Urokinase Plasminogen Activator in Complex with Its Receptor

The urokinase plasminogen activator binds to its cellular receptor with high affinity and initiates signaling cascades that are implicated in pathological processes including tumor growth, metastasis, and inflammation. We report the crystal structure at 1.9 angstroms of the urokinase receptor complexed with the urokinase amino-terminal fragment and an antibody against the receptor. The three domains of urokinase receptor form a concave shape with a central cone-shaped cavity where the urokinase fragment inserts. The structure provides insight into the flexibility of the urokinase receptor that enables its interaction with a wide variety of ligands and a basis for the design of urokinase-urokinase receptor antagonists.

黄明东博士,现为科学院福建物质结构研究所研究员,博士生导师,化学生物学研究室主任。1982年和1985年分别获得福州大学和厦门大学学士和硕士学位,之后任福建物质结构研究所助理研究员。1993年获得美国俄勒冈州立大学化学博士学位。随后在美国Scripps研究所和哈佛大学医学院工作。2002年应聘中国科学院****到福建物构所工作。有多年的从事生物学和化学的研究经验。他曾作为项目主持人和研究人员分别承担过美国心脏协会和多个美国国家卫生部(NIH)的科研项目。他的原创科研文章至2002年初被139种期刊引用394次。其中有三篇科研文章发表在国际的《科学》和《自然》类杂志上,其中一篇为第一作者。他的科研成果在许多国际会议上报告过,被其中四个国际会议上邀为大会口头报告,并被邀在许多科研单位交流报告过。社会活动方面,1997年创办美西福建促进会,任副会长;1996年创办美国圣地亚哥华夏中文学校,并任终身理事。2004年任福建生物工程协会副会长。他的主要研究方向为蛋白质的结构生物学研究。

Mingdong Huang, Ph.D, is currently Professor and Director of Chemical Biology Division of Fujian Institute of Research on the Structure of Matter (FIRSM), Chinese Academy of Sciences. He obtained his PhD in chemistry from Oregon State University (Corvallis, USA), and did his postdoctoral research with Dr. Ian A. Wilson at The Scripps Research Institute (San Diego, USA). His major research interest is on protein-protein interaction and protein-small molecule interaction. His research effort is currently funded by Hundred Talents Program of Chinese Academy of Sciences and State Key Project of National Science Foundation of China. His research works were published in peer-reviewed international journals, including Science and Nature.
