文章通讯作者是973计划重大项目“诱导多能干细胞的重编程机制”首席科学家王纲研究员,主要从事转录中介体与基因表达调控方面的研究。

 

脂肪细胞分化过程受到众多的信号通路和转录因子调节,其中一条十分重要的通路是Insulin信号通路,但Insulin通路的信号是如何传导至核内的转录网络还不十分清楚。在Insulin信号转导途径中,核内蛋白Krox20是已被发现的控制脂肪细胞分化过程的最早的转录因子,但Krox20如何被Insulin调控的具体机制也不清楚。

 

据生化所消息,王纲研究组的博士生汪炜和助研黄璐博士(共同第一作者)通过一系列的分子与细胞生物学实验,证明了Mediator复合物的Med23亚基和它的相互作用蛋白Elk1都是影响脂肪细胞分化的重要调节因子。这项研究发现,无论是敲除Med23或者Elk1,都会明显抑制脂肪细胞分化过程;Med23通过控制Krox20的基因转录水平来调节脂肪细胞分化;在Med23缺失的细胞中过表达Krox20能够挽救因Med23缺失而引起的分化缺陷。他们还通过进一步的生物化学实验具体分析了Med23控制Krox20的分子机制。

 

该工作揭示了Med23-Elk1的相互作用是连接Insulin信号通路与核内基因转录网络的重要节点,为脂肪的生成提供了新的分子解释,为干预肥胖及相关疾病提供了一种新的可能性。

 

该项工作得到国家科技部、中国科学院和上海市科委的经费支持。

Summary

Adipocyte differentiation is orchestrated by multiple signaling pathways and a temporally regulated transcriptional cascade. However, the mechanisms by which insulin signaling is linked to this cascade remain unclear. Here we show that the Med23 subunit of the Mediator Complex and its interacting transcription factor Elk1 are critical regulators of adipogenesis. Med23/ embryonic fibroblast cells were refractory to hormone-induced adipogenesis. Knockdown of either Med23 or Elk1, or overexpression of dominant-negative Elk1, inhibited adipogenesis. In the absence of either Elk1 or Med23, Krox20, an immediate early gene stimulated by insulin during adipogenesis, was uninducible. Moreover, the adipogenic defect in Med23-deficient cells was rescued by ectopic expression of Krox20 or one of its downstream factors, C/EBP or PPAR. Mechanistically, the insulin-stimulated, Med23-deficient preinitiation complex failed to initiate robust transcription of Krox20. Collectively, our results suggest that Med23 serves as a critical link transducing insulin signaling to the transcriptional cascade during adipocyte differentiation.

 

王纲简介:

1988年毕业于北京大学心理学系,获学士学位。

1990年至1993年获Greensboro Graduate Scholarship就读于University of North Carolina心理系,获硕士学位。1993年进入Tulane University攻读分子与细胞生物学博士,研究癌症相关基因的调控。曾两次荣获Student Research Award (From the Cancer Association of the Greater New Orleans), 一次获得First Place Poster Award (at the Oncology Update Meeting, New Orleans)

199812月从Tulane University获博士学位后,加入加州大学著名分子生物学家Arnold Berk的实验室,研究课题为Mediator与基因表达调控的功能关系。 在加州大学历任Postdoctoral Fellow, Research Associate III, & Assistant Researcher II。于20057月应聘回国加入中国科学院上海生命研究院生物化学及细胞生物学研究所,任研究员、博士生导师,中科院“****”获得者。

目前已获得“国家863项目”,“973重大基础研究项目子课题”,“上海市浦江人才计划”等资助。

 

研究工作

Mammalian Mediator是由大约30个蛋白质组成的多蛋白复合体(multiprotein complex, 是介于转录因子(transcription factor mRNA Polymerase II basal transcription machinery之间的联接桥梁。几乎所有的基因表达都需要Mediator的调节。不同的转录因子受到环境及发育信号的影响时,就会与Mediator中的某个特定的蛋白质发生相互作用,促进Polymerase IIGeneral Transcription Factors (GTFs) 在基因initiation sequence上的聚集并形成preinitiation complex (PIC), 进而调节基因表达的水平。

 

本实验室集中对Mediator的结构及功能作深入分析。过去我们对Mediator Complex的大小、数量、及非同一性(heterogeneity)作了分析;发现了Mediator 包含Sur2/TRAP100/TRAP95 submodule 揭示了腺病毒的E1A蛋白与Mediator在腺病毒感染与转化中的相互作用;同时还发现Mediator不仅能作为activatorPol II之间的桥梁,而且在召集Pol II之后,Mediator还会进一步刺激Pol II的活性,加速基因的表达。

 

我们未来的工作将继续应用蛋白化学、分子生物学与分子遗传学等方法,结合proteomicsgenomics的最新手段,研究在分子及细胞水平上Mediator可能参与的众多生物学事件,例如信号转导途径与基因表达的coupling、染色体结构的变化、 mRNA processing、在感染及转化中病毒基因的表达调控, 以及癌症的发生过程等等。
