浙大,首都医科大接连发表Nature子刊综述

生物通报道:英国著名杂志《Nature》是世界上最早的国际性科技期刊,其旗下的各大子刊也是各个领域中的顶级杂志,近期来自浙江大学,首都医科大学的研究人员分别在Nature子刊上发表综述,介绍了Hippo信号通路的新研究进展,以及高血压饮食相关综述。

在第一篇文章中,来自浙江大学生命研究院,以及加州大学圣地亚哥分校的研究人员结合近年来在相关领域的研究成果和最新进展,系统综述了Hippo信号传导通路组成及调控机制,并重点对器官大小和组织再生调控过程中细胞极性蛋白和细胞粘附蛋白对Hippo信号传导通路的影响及机制进行了综述和探讨。

生物体中组织器官大小和体积的调控一直是生物学研究的最基本的问题之一。Hippo信号通路是一条细胞抑制生长性信号通路,在进化过程中非常保守,多细胞动物果蝇、小鼠、哺乳动物中都存在Hippo信号通路。最早研究人员在果蝇中发现Hippo信号传导路径是调节细胞大小,器官体积的主要信号通路。之后研究人员证实Hippo信号通路具有高度保守型,果蝇中Hippo信号通路的成员都能在高等生物中找到对应的同源物。在哺乳动物中,Hippo信号通路通路上游的膜蛋白受体感受到胞外的生长抑制信号后,经过一系列激酶复合物的磷酸化级联反应,最终将磷酸化下游的效应因子YAP。磷酸化的YAP与细胞骨架蛋白相互作用,被滞留在胞质内,不能进入细胞核行使其转录激活功能,从而实现对器官大小和体积的调控。此外,近年来的研究证实Hippo信号传导通路还在癌症发生、组织再生以及干细胞的功能调控上发挥重要的作用。

近年来关于Hippo信号传导通路的研究十分活跃,处于国际科技发展的前沿。浙江大学生命科学研究院的多个课题组对于调控Hippo信号传导通路同时在展开深入性的研究。不久前浙江大学生命科学研究院的黄俊教授在对hippo信号通路关键蛋白:YAP1的研究中取得突破性进展。研究人员克隆出了两个YAP1结合蛋白AMOTL1和AMOTL2,并发现它们通过影响YAP1的核质穿梭副调控YAP的功能。进一步的研究发现,在MCF10A细胞中降低AMOTL2的表达能够促进细胞发生上皮细胞向间充质细胞的转化(EMT),而这一表型恰好与YAP1在该细胞中过量表达的表型一致。这一研究成果发表在美国生化与分子生物学学会主办的JBC杂志上。

另外一篇文章中,来自首都医科大学附属安贞医院,北京心肺血管疾病研究所的研究人员发表了题为“Dietary factors associated with hypertension”的文章,解析了与高血压有关的一些饮食因素,比较了各国和不同组织提出的预防指导措施,为高血压的基础研究,和临床研究提供了重要的资料。

高血压是心脑血管疾病最重要的危险因素之一,目前我国高血压患者超过2亿,高血压已是我国人群最重要的心血管病危险因素,如何控制高血压病是我国乃至全世界疾病控制所面临的挑战。

随着医学研究的不断发展,医学界对高血压病的认识不断深入,包括病因、发病机制、病理生理和预后等,研究证明饮食与高血压密切相关,虽然遗传因素决定了很大的一部分,但是我们每天吃的食物会影响到身体的各个器官,包括动脉,血管。

在这篇综述中,研究人员总结了影响血压和高血压的最新饮食因素,比较了各国和不同组织提出的高血压患者的推荐饮食类型和摄入量,以及其高血压预防指导手册,从中概括了高血压控制的全球健康饮食策略。

研究人员还评估了27个饮食因素,认为其中的17个具有针对高血压的保护性作用,6个是高血压风险因素,而其余的几个因素对于血压的影响尚无定论。文章还指出过量的钠摄入会引发高血压,很多证据也证明水果,蔬菜,低脂乳制品,和低钠,低饱和脂肪酸则是预防和降低高血压的积极饮食,值得注意的是各高血压控制和预防指导中的推荐饮食类型,和摄入量都存在显著差异。

The Hippo pathway in organ size control, tissue regeneration and stem cell self-renewal

Precise control of organ size is crucial during animal development and regeneration. In Drosophila and mammals, studies over the past decade have uncovered a critical role for the Hippo tumour-suppressor pathway in the regulation of organ size. Dysregulation of this pathway leads to massive overgrowth of tissue. The Hippo signalling pathway is highly conserved and limits organ size by phosphorylating and inhibiting the transcription co-activators YAP and TAZ in mammals and Yki in Drosophila, key regulators of proliferation and apoptosis. The Hippo pathway also has a critical role in the self-renewal and expansion of stem cells and tissue-specific progenitor cells, and has important functions in tissue regeneration. Emerging evidence shows that the Hippo pathway is regulated by cell polarity, cell adhesion and cell junction proteins. In this review we summarize current understanding of the composition and regulation of the Hippo pathway, and discuss how cell polarity and cell adhesion proteins inform the role of this pathway in organ size control and regeneration.

Dietary factors associated with hypertension

Hypertension is one of the major risk factors for cardiovascular disease, with an impact on global health. Multiple studies have suggested that various dietary factors are associated with blood pressure (BP) and hypertension. The purpose of this Review is to provide up-to-date knowledge on the impact of dietary factors on BP and hypertension, to compare types and recommended intakes of dietary factors in hypertension management and prevention guidelines from different countries and organizations, and to outline global population-based healthy-diet strategies for hypertension control. Of the 27 dietary factors we evaluated on the basis of specified review criteria, 17 have been proposed to have protective effects against hypertension, six were proposed to be risk factors for hypertension, and the association between BP and the remaining factors was considered inconclusive. Excessive sodium intake is a causal risk factor for hypertension, whereas a diet rich in fruit, vegetables, and low-fat dairy products, and low in sodium and saturated fat has been recommended to prevent and reduce hypertension on the basis of strong evidence. Notable differences exist in the recommended types and intakes of dietary factors among available hypertension management and prevention guidelines. Available evidence supports the vigorous implementation of dietary strategies against hypertension through population-based, national action plans.
