Takara公司推出的Capturem™ Extracellular Vesicle Isolation Kit (Mini)（Code No. 635741）可在30 min内实现血浆等多种生物体液样品中细胞外囊泡（EVs）的小量快速提取，产物纯度高、污染少。

　　

细胞外囊泡(EVs)例如外泌体，虽然体积很小，但在正常的生理过程(如免疫应答、神经元功能和干细胞维持)和疾病(如癌症和肝病) 的发生发展中发挥着重要作用。EVs研究的一个关键瓶颈就是囊泡的分离，传统的做法是通过超速离心法实现的，但它耗时长，需要特殊的仪器，而且在高速离心的过程中可能会损伤囊泡，收量也很低。近年来，研究人员使用试剂沉淀法来简化EVs的提取操作流程，但是基于沉淀的技术往往不一致，产量低且纯度低。因此，急需一种方法能在保证纯度和收量的前提下，加快EVs提取的操作流程。

Takara公司推出的Capturem™ Extracellular Vesicle Isolation Kit (Mini)（Code No. 635741）有效地解决了这些问题。该产品采用了一种新型的膜技术——Capturem™膜，将可以与EVs特异性结合的凝集素（非抗体）固定在膜上，以选择性地分离EVs。Capturem™膜经过化学修饰增加了膜孔表面积，从而实现了更高的结合能力，可以分离得到高纯度的浓缩样品。此外，Capturem™膜被组装到离心柱中，使用普通的台式离心机，即可在30分钟内实现EVs的快速分离。

Western blot characterization of exosome preparations by Capturem kits and ultracentrifugation (UC). Western blotting shows that the Capturem kit contains higher levels of exosome-specific markers CD63, CD9, and Alix and lower levels of the non-exosomal protein calnexin and carryover protein albumin. In contrast, the sample prepared using UC contains considerably higher levels of albumin and calnexin. Each lane was loaded with 5μg of total protein as measured using the Pierce BCA Protein Assay Kit (Thermo Fisher Scientific).
